Glycosaminoglycans in Human Neutrophils and Leukemic
نویسندگان
چکیده
Chondroitin sulfate is known to be present in normal and leukemic myeloid cells; however. its definitive subcellular location and association with other glycosaminoglycans (GAGs) has not been demonstrated. We have studied the type and distribution of GAGs in neutrophil granule subpopulations of normal and leukemic myeloid cells using ultrastructural. cytochemical. immunologic. and biochemical methods. At the ultrastructural level. high-iron diaminethiocarbohydrazide-silver proteinate (HID-TCH-SP) stained sulfated glycoconjugates selectively in immature primary granules of normal promyelocytes and Auer rods and immature granules of leukemic myeloblasts. Staining was weak or absent in mature primary granules. whereas tertiary granules stained moderately. Primary granule staining with HID-TCH-SP was greatly diminished by prior treatment of the specimens with chondroitinase ABC and/ or nitrous acid. indicating the presence of chondroitin sulfate and N-sulfated glycosaminoglycan. Immunostaining of myeloid cells with a rabbit antichondroitin 4-sulfate and

متن کاملSensitivity of Human Leukemic Cells to Carvedilol
Background and Objective: Carvedilol, a non-specific β-blocker, has been used for treatment of hypertension, stroke and congestive heart failure. The therapeutic effects of β blockers in cancer patients have been shown. Carvedilol has considerable anti-inflammatory, anti-tumor and anti-angiogenic properties. In this study, the effects of carvedilol on proliferation of human U937 and Molt-4 leuk...
متن کاملExpression Pattern of Interferon-γ in Human Leukemic T Cell Lines Following Treatment with Phytoheamagglutinin, phorbol myristate acetate and Lipopolysaccharide
Background: As a T helper type 1 (Th1) derived cytokine, Interferon gamma (IFN-γ) is an important regulator of inflammatory immune responses. Furthermore, IFN-γ plays an essential role in defense against tumors and intracellular pathogens. This study was designed to assess the pattern of IFN-γ production in human leukemic (Jurkat and Molt-4) T cell lines in vitro. Methods: Ju...
متن کاملAssessment of Cytotoxicity of Dimethyl Sulfoxide in Human Hematopoietic Tumor Cell Lines
Background: Dimethyl Sulfoxide (DMSO) is a solvent most broadly used as a cryopreservative agent. Antitumor effects of DMSO is a recently recognized phenomenon. In this study, cytotoxic effects of DMSO on human monocytes and T leukemic cell lines has been investigated in vitro. Methods: Human leukemic T cells (Molt-4 and Jurkat) and monocytes (U937 and THP1) were cultured in complete RPMI medi...
